To make it real simple Get a ratio cup it has all the markings on the cup for pre mix and also has CCs so u can measure your crankcase oil. It is fool proof. I take the empty 1 liter bottle and measure for six gallons then i marked the bottle so i can refill. The ratio cup gos up to 2 1/2 gallons It is only $4 Worth every penny

To answer the question directly:
32:1 is 32 parts gas to 1 part oil. So if you use 32 oz of gas you need 1 oz of oil. If you use 32 gallons of gas, you need 1 gallon of oil.
ie. 32:1, 40:1 etc are just ratios of gas to oil.
1 US gallon = 128 oz
Therefore, for a 32:1 mix:
32 oz gas needs 1 oz oil
128oz gas needs 4 oz oil
or 1 gallon of gas needs 4 oz of oil
or 5 gallons of gas needs 20 oz of oil
Once you figure out the amount of oil required, write it on your gas cans as amount of oil per gallon. ie. I usually write "32:1 = 4oz/gallon". This way it is easy to multiply by the number of gallons you are putting in.
In US gallons:
20:1 = 6.4oz/gallon
32:1 = 4oz/gallon
40:1 = 3.2oz/gallon
