92 Trail boss 250 2x4 Hard Starting

Hi all, new to forum.
Having problem with 92 Trail Boss 250 2x4. When cold (Sitting overnight) it won't start Choke or No choke - Pulled plug and its completely dry. Good Spark. I rebuilt the Carb and it started fine - Next day wouldn't start again.
Tried putting a little gas in from the air cleaner side - No start
Pulled the thottle assy out and put a little gas in here and it fires right up - Runs for the day with no problem.
Sitting here typing this i realized that maybe i should check to see if the manual choke is working properly.
If it is , what should i check next.
Can i check the intake boot by spraying alittle starting fluid to see if it sucks it in through a crack?
I don't have a compression gauge to check compression.
What am i missing??

If dont have compression tester,can use old timey method: pull plug,place thumb on sparkplug hole,hit starter,IF enough compression to blow thumb up,possible good enough to crank! I do this on pwc's on a regular basis,just to see if one cylinder low or holed. Compression,Fuel,Fire,and Air, gotta be one or the other. Good luck

Update - I removed the Choke cable to make sure that it was working.
I sprayed Card cleaner through the port and hit the starter and it fired.
I then re-installed the choke cable and hit the starter and nothing. Adjusted the cable up and it fired off. Hopefully all it needed was to be adjusted some.
When i get home from work tonight i will see what happens when it sits over night.
I'll also do the old thumb trick to check compression tonight.
Talking to my brother last night, the bike has never had any work done other than normal maintenance and replacement items. He bought it in 92 ran it for 8 years, then let his sons run it for 6 years and then i bought it from him for my kids and i have had it 2 years. It will not suprise me if the compression would be low after 16 years.

Would not start tonight - Borrowed a compression gauge and i got 105 & 108 psi so i'm good in this area.
I replaced the spark plug and no start.
I removed the seat and top of air cleaner, covered the round tube with my hand and it started. i ran it maybe 2 minutes.
Is this telling me its a choke adjustment problem?
I adjusted the choke up another 1-2 turns and left it sit for a hour and half and it started ok. When warmed up it runs great, breaks the tires loose and goes maybe 35 mph.
I will check it first thing in the morning to see what happens.
Am i doing the right toubleshooting?
