With these things start proceedure is everything. First off turn the choke on and DO NOT touch the throttle. If you do it won't start. No crank the engine and still don't touch the throttle. It will start and run with out the throttle when cold. When warn crank the engine and then just a touch of throttle. FYI, no accerlerator pumps on these carbs.

You might also want to check your intake and exhaust valves. They may be out of adjustment which can cause hard starting.
Also, check that your spark plug is the correct heat range, my manual says your machine should should take a NGK DR7EA spark plug(gapped at 0.6-0.7mm). You could try a little hotter plug, but I am not familiar with NGK plugs enough to say wether a NGK DR6EA or a NGK DR8EA plug would be hotter.
As Bear mentioned, find the machines starting procedure. What ever it is, and follow it and starting will be a lot easier.
I have actually found that my 400 starts easier with the choke off on the first try and then choking it the second try if it fires and dies. While my TRV is the exact opposite.

Sounds like your enrichening system (Takes the place of a choke by dumping extra fuel behind the slide) isn't working due to being plugged or the cable broke or frozen ot the valve stuck or passages plugged.

It has a lever, labeled choke, but the cable operates an enrichment valve. There is no choke butterfly on this model. There is a slide and a throttle valve but no choke betterfly valve. Check here for the carb breakdown:
