I believe you are talking about the diaphram that has the needle jet in it? I believe it is operated off a vacum from the carb and acts like a fuel pump on the older models. So if it is torn or leaking I would suspect that the engine would run lean and or be hard to start due to lack of fuel.
It has been my experience that you can usually see a tear or cracking on the diaphram. Plus it can be a real pain to put back together. If it doesn't seat correctly when assembled then it will be like having a hole in it.
What symtoms is the engine doing that you suspect its TU?

My quad has all of a sudden gotten a serious rich condition,I have tried several different combinations to resolve this but dont get it.I have about ready to throw the towel in on AC products.I've been stung a few 2 many times.
I have cleaned the carb(multiple) times,inline fuel filter,new air filter,just gettin frustrated.I am a maintenance tech so i hate having to concede and take it to somebody else but my head is scratched to the skull bone.
My carb has
142.5 main
40 pilot
5d40 needle
e clip in second slot down from top on needle
could my needle be worn?

You might also check the mixture screw. I think it should be about 1 3/4 turn out. I usually set mine there after I clean the carb and it does fine. If it is out or in too far then it will cause issues at high or low rpm's.
Also, I think your 454 is now a the 500 AC engine? If so I think they are now using a 138 main jet (at least that is what my 500TRV has). You may also want to check your radiator as overeating or over cooling will cause some issues as well.
