Don't put it away dirty!!!
Dirt and mud hold moisture, make sure you clean it thoroughly after each ride.
Make sure you keep the air filter clean, do this about once a month, depending on how often you ride.
Change the oil on a regular basis, exspecially since the 250EX does not have an oil filter, just a screen.

It depends on if you ride in sand, mud, dust, or maybe a clean trail. The following is what I do. Of course, you don't have a chain.
Here's what I do everytime:
Wash it
Armor or lemon pledge all the plastic
Adjust chain & lube
Check the tires (also check for tears & slices)
Check the oil
Check the plug
Then there's the other stuff I do about every 20 hours:
Change the oil & filter
Clean & oil the air filter
Lube the cables
Check the brakes
Adjust the parking brake
Generally inspect all bolts & nuts

This is the most maintenance free quad on the planet!
Keep the air filter cleaned, change the oil every 20 hours or so, adjust the valves every 100 hours, and change the oil in the rear drive every season.

Rear drive takes hypoid gear oil. Only takes a few ounces. I actually change mine every spring and fall.
Not sure exactly what the owner's manual says, but I think either GL4 or GL5 is fine, but it must be "hypoid" and around 80W as I recall.
I use Maxima synthetic 75W90: http://www.maximausa.com/produ...ubes/synthgearlube.asp
Honda's own "Shaft Drive Oil", is hypoid 80W90, and GL4/GL5.

Rear drive takes hypoid gear oil. Only takes a few ounces. I actually change mine every spring and fall.

It's going to get confusing, becasue Honda now calls the 250EX...the 250X!!!
