IMO no. The 400ex already revs plenty high. Higher than I would ever rev mine. I never hit the rev limiter in the 400ex drag racing and the only time I ever did doing hill shoots was once I topped over a hill there was a ditch on the other side that I didn't quite miss as planned. I buried the front wheels and went to fly over the handle bars pinning my knee against the throttle. I had the mind set to be holding the clutch at the time but, the rpm's pegged like crazy. No way would I feel safe having my quad rev higher than that in any circumstances.

My neighbor has a couple 400ex's and he has Rev boxes on both of them. He swears by them. With a Rev box it's not just an RPM increase, it's also a timing advance which adds a little HP, which the EX could always use more of.

True, albeit ever so slight hp increase according to dyno's but, ever lil' bit helps. So if you already have pipe/filter/jet. It may add a bit of performance. Personally before rev box I'd then get rims and tires, they reduce weight and are stronger. Then some nerfs, good grab bar, maybe handle bars... Just a personal preference I guess. To me rev box would be one the last things to spend my cash on.
