I'm no pro on dirt and have never been on sand, but i can offer a little....
What size/type of tires are you gonna run? The paddles will hurt your mph on dirt.
Are you gonna run 300ft or 600ft?
Are you running an aftermarket cdi?
With your gearing and a 20" tire, you'll hit the limiter just before the 600ft mark unless you have a higher than stk limit.
With your gearing, i would take off in 2nd. Some say they let the clutch go in first and immediatly shift to second and do well. I did better leaving in 2nd.

With the 22" tires 15/42 should be perfect!!
Are you running with a wheelie bar? If not, do yourself a favor and go get two pieces of 1" black iron water pipe, cut 18" long and drill holes 17" center to center to replace front shocks so you can dump the clutch and get a much better holeshot.
I run 22" 4-Snows with 15/40 gearing and ALWAYS take off in 2nd.
