"I did an experiment tonight. I flatted the rear tire on the 400EX (Holeshots), and then refilled it. One 16 gram BB cartridge was not enough to have a rideable tire. Two cartridges got it up to a rideable 3.5 lbs. And , a third got it up to 6 lbs. In addition, you are going to need at least one cartridge to initially inflate the tire to find the leak and plug it.
So, I figure 4 cartridges minimum for a sport bike rear tire. Another way to go might be the "monster air chuck" and 45 gram cylinders. See- http://www.innovationsaz.com/

I carry the 25 gram threaded cartridges plus a controlable inflator top for it, two of those is more than enough to fill up my 26" mud tires to rideable pressure. But as reconranger mentioned you will may need to use some to find the puncture. I carry about 10 with me all the time.

Keep shopping around, I found a box of 50 at sportmans warehouse for 6 bucks. Those co2 cartridges probably can be found even cheaper online.
I agree... one to inflate enough to find the leak, plug tire, and another will get it to the point where you could ride it, but the third will get it to about 4 lbs.
