hey welcome to the forum!
look for an air leak (check seals, gaskets, carb boot, etc), WEAK spark (what color do you have now cause this is a common issue), leaky/broken reeds, carb adjustments or gunk in the carb.
check them all out its very possible that one of them is your problem.
also try towing the bike around in 3rd, VERY good chances it will start. and thats still an unresolved phenomena that plagues many of us zuki 250r riders. it simply wont kickstart but yet it has perfect compression, ignition, and carburation and yet it only starts with a good tug. and after that it runs PERFECT and will kickstart back up until it cools off. im a mechanic by trade and it has boggled my mind for the past year or so.
